For most visitors to Florence, time is the enemy. Perhaps the majority of visitors stay no longer than 2 days, many struggle to find a day in their itinerary.
For the mainstream first time visitor to Florence, there are 4 "must see" sights in the city that you can just about do justice to, in one full day.

For longer stays these four attractions will form the backbone of your visit. If you have a keen interest in art and architecture you'll need a minimum of 2 days.
The Duomo (cathedral) and Ponte Vecchio are the two landmark attractions that will make even the most travelled reach for their camera.
These two landmarks are supported by two blockbuster galleries.
The Uffizi Gallery is one of the world's finest and after the Vatican Museum is the most visited museum/gallery in Italy.
The Accademia Gallery is much smaller but has the show stopping exhibit of the world famous Michelangelo's Statue of David.
For the mainstream visitor around 90 minutes at each is probably going to be enough, for the person with a real interest in art set aside at least 3-4 hours for the Uffizi.
All four attractions are in the historic centre of Florence, you can walk between all four in 20 minutes, and walking is really your only viable way of getting around the centre.
Queues for the Uffizi Gallery and Accademia Gallery are very long, you really do need to purchase tickets in advance if visiting independently.
